cylinder won't lock

My Taurus model 85 5 shot 38 spec. will not lock the cylinder in place for discharge.  I've had it apart and nothing appears broken.  What makes the cylinder stop pop up into its notch?   The screw just above the trigger that holds the sideplate fell out and the cylinder fell out but that has been replaced.  Help!!!!

Below is a link to a diagram.. What screw fell out and what do you mean the cylinder won't lock into place?? The cylinder won't lock into the frame open or the cylinder won't lock into alignment with the barrel??

Gunnut They cylinder closes and locks in the frame ok but the cylinder will not lock in proper chamber alignment  for the gun to shoot.  the cylinder stop does not pop up out of the frame into its notch in the cylinder.  the yoke and cylinder assembly fell out when the front screw that holds in the sideplate and retains the yoke in the frame came out.

It's really hard to give the eact cause the weapon isn't working.. Look at the driagram and notice part #20 the bolt.. It locks the cylinder into position by popping thru the bottom of the cylinder opening in the frame.. Perhaps a look at the functioning of the piece with the sideplate off would give an idea of the problem. It could be many things, broken parts/springs, burrs...??? Remove the sideplate and cycle the action slowly and watch the parts' functions..

Had a similar problem. Try removing the cylinder bolt and polishing both sides and the slot through which it passes. Some taurus parts seem to have some rough edges and like to bind or hang up. This cured the problem in my gun.
