The 'perfect cartridge", or caliber, is in the mind of the user.  For rifle the most popular is 30 caliber by far, and for good reason.  It doesn't take a truckload of lead to load up a box of ammo, the bullets are easy to handle and the standard rifing twists for the various cartridges seem to be very compatable with lead bullets.  In other words, it seems that results tend to 'just happen' most often when the shooter has a 30 cal.
A good general  consensus seems to be that larger bores are easier with cast than smaller, and I've found that the larger the bore the faster one can push plainbase without leading.

In handguns, I don't know of any that are bad in calibers from 32 and larger, or superior, if accuracy is the main concern.  Of coarse large calibers will flatten larger animals easier.  My favorite is the 357 in a four inch revolver simply because it's so easy to carry, when working, hiking or hunting.  I don't worry about big stuff getting me even though I know I'm under armed for grizzly, which do live in my area, so my notions on caliber are personal and I'm not trying to put them on anyone else.

A bit about personal ideas forming what we think is best.  -- A friend up here was once hunting bear with hounds, when a big one started to get away at breakneck speed.  He yelled at the hunters with him, "He's gonna get away!" and snapped off a shot with his 357, loaded with a 125 grain pointy nosed SWC, which hit the bear and dropped it in it's tracks.  The next week end, every one of the guys in his party showed up with S&W 357 revolvers!         Their beliefs in the gun were formed from an accidental 'good shot'!

Veral,
Your remarks on personal ideas being formed [by experiences] are right on target.
It is always interesting to have others tell what they think is best [about anything] and to offer support for their thinking if they have any. (Many seem unable to explain why they think anything at all.)
Most of us will have our preferences evolve over time as a direct result of observations, experiences and lessons learned along the way. We should always be prepared to modify our thinking when new information comes along. In theory, the day we fail to learn something should be the day our lease on life expires.

My personal "favorites" regarding all shooting subjects have gone through many changes, (some in a near full-circle,) from boyhood to present, (approximately 50 years.)

45-70.gov,
I have a few ideas about the "perfect" [all-purpose] firearm-cartridge & rifle-handgun combination but it would take a complete essay on each facet of that subject to get the whole thought explained.
I'll offer this teaser, it is 45 caliber, (well actually .460 caliber.)
While I agree completely with Veral's remarks about the 30 caliber, (and if we were talking about rifles only that caliber would be tough to beat,) I now am prepared to argue in favor of the 45/70 in a lever action (Marlin) with several bells and whistles that are not factory-offered along with a revolver that shoots the same-caliber in a cartridge that is made from a shortened 45/70 case, (currently only available by custom gunsmiths.)
Cursory argument:
This rifle and revolver combination can/could be loaded for any use from realistic squirrel & rabbit [small game] shooting to the biggest, toughest animals on earth giving performance ranging from adequate to excellent in all circumstances.
A whole book could be written to support this opinion from what I've personally seen demonstrated or learned along with information that comes via several avenues.
"Stop global whining!"

I think I just deleted a post by 45-70 that I shouldn't have.  I thought your first post had accidently got posted a second time, then after hitting the bomb button, read part of a question I hadn't seen in your first post!   I'm sorry.  Hang it up for another look see, please.

The question was about 44 caliber, but that's all I saw. -- Without knowing what your question was, I'll say it is the most do it all caliber there is when a person has a rifle and revolver to chamber it.  The rifle loads will take down anything on the north american continent as efficiently as a 300 magnum rifle.  And it can be loaded easily to power levels that make shooting painful with just a few shots.  Of coarse the gun is far lighter than a 45-70 lever locker, which means the larger cartridge gun can deliver more power before recoil get miserable, but it's power American hunters have no need for.  So, in my very opinionated mind, why carry more weight around than one needs.  I'm such a nerd on this that I cut the magazine on a Marlin 30-30 to hold just two rounds, and trimed the wood to minimum size.  Very nice little rifle now, which I've never hunted with!  But it's a gun I like a lot.  When lever gun magazines are filled, poi changes as they are shot down, which is something I don't like.  Also, since I have never shot more than twice at a game animal in my 50 + years of hunting, I see no reason to carry more than three rounds in the gun, which I am carrying IN MY HAND.  I prefer to carry my extra rounds in a pocket.

What I've just said about the 44 holds true for the 41 also, which I've never used in either type of gun, and if one isn't going to hunt game larger than elk, a 357 will get the job done with stunning efficiency if my 180 gr FN is used, whizzing along at about 1800 fps. -- My wife loves to hunt, but doesn't get too technical about which gun/ caliber she is carrying.  She calls the 357 "that quite gun".  It's her favorite and shes used most everything up to 30-06.  She likes it because it's bark doesn't bite, and it doesn't hurt her shoulder, yet it kills like lightening.  She has taken one elk, a coyote, at 200 yards running full out, several deer and quite a few smaller critters with it.
