I want the 45 for flatness. If you cant kill a deer with a 45 cal. bullet then I'll quit hunting. Especially since you have about 350fps. more with the 45 cal. over the 50. Foot pounds of energy at 100yds are about the same as the 50 cal.

Yep, I'll second Randy on the 50 cal. the beauty is you can get 45 cal performance out the 50 by using Dead Center Bullets from Cecil Epp. http://www.prbullet.com/pts.htm

You deserve a better answer.

Whether Encore or Omega, the barrel is not tapered. A .45 caliber gun gets you a smaller hole in the same pipe. That means more weight, and more nose-heaviness. It also give you less barrel volume / combustion space.

With a .50 caliber, you have a .50 with conicals, or a .45 / .44 / .40 / .357 gun with sabots. The fifty is easily a .40 caliber rifle, but the .45 can never be a .50 cal for larger game. And, those .40 cal Dead Centers shoot great out of the .50 caliber Thompsons.
