BTW You can get an early idea of the trajectories by comparing the 44-40 and 44 magnum in rifle calibers. Remington, for example, has a nice presentation of ballistics for their cartridges. The link here is for just these two:

The key here is that with both sighted in at 100 yd, the difference in drop at 150 yards is only about 5 inches. This means that DennyRoark's suggested sight in is an excellent starting point.
Nonetheless, you will be well served by going through the steps of getting your MV, BC, and trajectory, and confirming/adjusting your points of impact at the range.
