I recently got me a Marlin Cowboy with the Marble's semi-buckhorn rear sight.  This type of sight is new to me.  What sort of alignment should I be looking at (that is, where should the front dot be sitting in relation to the rear opening, white diamond, etc.  ... besides "in front" :) )  Thanks in advance,  Peter F.

Hey Peter

I have two Marlins and what works for me is having the front bead "sitting" on the top of the rear sight.  Now that's not the way I was taught but seems to be the way I use the Marlin sights "naturally."  Seems to hit pretty much where I point it, too.

p.s. remember for CAS the white diamond needs to be blacked out--I used a magic marker to do the job on mine.

Yep, for SASS you need to black out the diamond. Or, you can reverse the insert in the rear sight so the diamond faces away from the shooter.
