difference in 22LR only chamber vs 22 S,L,LR chamber?

Is there a physical difference in the chambers of 22s that shoot Shorts and Longs versus the rifles that are rated for Long Rifle only?
Or is it the action that determines it, i.e, won't cycle or feed right?

You got it - the action is the determining factor.

When it's stamped 22 S,L,or LR it means the gun will function/feed with all three types.  If it's stamped 22LR only it means it will only feed properly or operate the action with 22 LR ammo.

For example the Ruger 10/22 is designed to operate with 22LR ammo (to fully cycle the action), but you could fire 22 shorts or longs in it by manually loading one round at a time - turning it into a really awkward singleshot!

So it should not hurt the chamber of my CZ 452 if I single loaded shorts in it, for hours of basement plinking fun?

  If you shoot a lot of them, you may have to clean the chamber...  That's about it...

  CCI CB longs would be an even better choise for what you want to do...
