Back off on the power ring a little, IE: 8.5x instead of 9x or 3.5x instead of 3x, I've read that on some scopes leaving the power ring adjustment at the end of travel can lock windage and elevation, worth a try.  ;)

Nice recycling there, I like the new 'safety' targets...........
IME, any scope that exhibits 'knob/adjusting issues' is suspect at best. Fortunately I have a couple of old El Paso Weavers that still are my dependables for if anything else is in question (or open sights; what a novel idea....).

I use H110 and TB in my 45 Colt loads. A couple of things I have learned. The chamber on my BCC is generous shall we say. I do not full length resize - only neck size to get enough neck tension to seat the bullets and use a strong crimp. I seat longer with TB and shorter with H110. If I seat longer with H110 I get sooting. Mine shoots well with either cast or jacketed but in cast I use a GC 300 gr fairly hard bullet from Beartooth and it wants a mid to high charge of 110 (using Ruger only data). With anything less than @ 85% of Max, I get poor accuracy and sooting.

It may be worth just to ream to 454, as mine is. It strikes me that everyone that has done so is getting better results even in 45 Colt.

Have you tried cast bullets of a dia. that does not require even neck sizing, just seating with a tiny bit of neck tension and a slight crimp?

No, my bullets are sized anywhere between .452 and .454. Normally they all shoot very well. I still haven't even slugged my barrel yet. ::)

I just checked my Colt barrel, it will chamber 405gr .460" cast!  ;D

The fit I refer to has nothing to do with slugging the barrel, just fitting the bullet to the fully fire-formed case mouth. After you get that right it WILL swage to the barrel just fine on its own.

 I just tried a few different bullets in fired brass. The best fit comes from a 300gr bullet that measures .454". It chambers with no issues.

With my lighter Unique loads, I find the Sierra Sportsmaster 240 gr. HP to perform great at sub 1200 FPS if you want a bullet that is accurate and does well on the low end.  I took my buck this last year with that bullet and he went about 20 yards.  First bullet went through his chest broadside and penetrated both heart ventricles, so it couldn't take long.  I ended up taking him with my Blackhawk, because I couldn't get the carbine up without notice.  Was glad I had the pistola ready.  ;)  The faster H110 loads and Reloader7 loads I use I put the 300 gr. XTP Magnum into it's place.  I took last years buck with this combo out of the colt and he ran no further.  I hit him at a full run a little far back, but still put him down in about 10 leaps.  ;)  As soon as I get the reloading center set up after this move, I've been given about 50 300 gr. Lee Molded bullets from wheel weights.  Wanting to see how they do in my carbine/blackhawk.  8)

As previously reported, I think my rifle's problem is not the rifle at all. I changed to a Burris Mini 4X and got outstanding results. I sighted it at 25, 50, and then 100yds. With just a few adjustments of the scope I was able to touch holes at 100yds. This old (1983) scope is lightweight, short and still clear and concise. Problem Solved!

Next I plan to load some of the 300gr boolits I received from srussel and bikerbeans. I think a little less weight and a bit more velocity will work quite well. My current load is 16.5gr 2400 and the .452" 335gr Cast Performance WFLGC.

What's your opinions on these Magnum XTP bummers in the Long Colt.

I bought some of those Magnum 300 XTP bullets before I had a Cassull thi king it might be better.  I have shot two maybe three deer with it and my 20+g 296 load and altho they work perfectly with game was recovered quickly and thru and thru penetration. Previously I used Speer 300 plated Sp with the same loading and once I shot end to end thru a doe. So much for needing a tougher bullet. ::)

I use te magnums in the Cassull and stick with my WFN cast and Speer/Sierra 300g bullets in the Colt. This is for both rifle and pistol.

If you didn't completely remove the iron sights, I'd drop up to 25 yards, do a 5 shot group and see what you get with the iron sights for a group.  If you get consistency with that you can pretty much be sure you are getting some issue with your scope.  I am trying out the Ironsighter 1 piece see through base this week on my superlite .223.  Looks like a nice solid base and lets me still see the irons for close in shots.  ;)
