It won't hurt anything.  It's a common practice in heavy-recoiling handguns.

not a problem.
i've put maybe a hundred
through mine.
i've mostly shot the
regular magnum rounds though.

Now ya need to rent the .445 reamer.  It's a super special Handi round.  You can still shoot all the others and have a big bad boy too.

445 Supermag was a Dan Wesson revolver chambering, it does for the 44 magnum what the 357 Maximum does for the 357 Mag either are simple hand rechambers, 44 Russian, 44Spl, 44mag and a couple others can all be shot in it. 300gr bullets to around 1900-2000fps are normal from a rifle.

I too have a 44 mag handi. I plan on reaming mine to 445 super mag. Your handi rifle needs to have a sb2 receiver if you plan on reaming it to 445super mag which you can read about in the FAQ  Heres a great link about the 445 from members here from a few years back  http://www.gboreloaded.com/forums/hr-centerfire-rifles/445-super-mag-223050/
DON’T get in a gun fight with a buffalo hunter there is no such thing as cover against a 45 70
