In my TC Omega I like the .44 cal/ 300 gr Keith Nose Hollowpoint. 

In a centerfire rifle I really like my .35 whelen.

Child of the 30-06, father of the 308.....
the 300 Savage is more than adaquate for taking Maine whitetails without punching your shoulder. This cartridge in a Featherweight 99, does the job fine, if I do my part.

I'm a creature of habit with what works for me ... 30-30 and 30-06. Guess that's why I've got 3 rifles of one and 2 of the other and danged it may get more ;D

I have to agree with .308 sniper, I've used 270's, 30-06's, and now 308. The 308 is my favorite so far ;D

I'm still hunting for a "favorite" or "best" deer rifle.  That's why I buy a couple a year and work 'em up.  So far, every one I"ve tried has done well.

I've taken deer with the .30/30, .270, .308 and .30/06.  I know there are a lot of folks who like the .270 and maybe it was the Core-Lokt bullet I was using but I wasn't satisfied, regardless.  The .30/30 and the .308 have lain down deer and I mean RIGHT NOW!  The .30/30 with 150 gr. Winchester hollow points and the .308 with regular old Winchester Power Points have worked the best for me.  One gun?  .308 Winchester.  Ballistic twin of the .30/06 when bullets of less than 180 grains are used, light recoil and it's very accurate.

Poachers kill deer with .22. that being said if you want to consistantly kill deer, practice your shooting, and unless you are wealthy the mega wiz bang magnums are mostly overkill, hard to find and expensive.
And if you practice, shoot the least expensive, but effective round.  And one that dosen't kick hell out of you.  That leaves 3006, 308 and 270.  That ammo is always on sale.  Even cheaper to reload.  check out the price for brass.  And I don't think anyone will fault those choices for any deer hunt in US.

I have used a single shot 280 exclussively since 1985, until this year I am now using a Browning A bolt in of cousre 280.  This caliber is more popular than people think.

I'm careful with my shots and it's not often that I don't fill my tag during deer season. Not sure how I can do much better than that.  I respect the guys who use other rigs, but for me, I don't feel like a deer-hunter unless I have a .30-30 cradled over my arm when I'm deep in the pine woods.

The answer to this BEST question is really one of location:
 I have hunted 3 geographical US locations.  Nevada:  shots 100-300+ yds.  Need a flat shooter, 25.06/270/280/3006/7mm.mag/

S. Louisiana:  most shots under 30yd. in heavy brush, deer small, Buckshot works great.

Midwest: Kansas,  most shots 75-150yd.  .308 is perfect, but anything that will kill clean works.  I have used 3006/7.5x55/762.x39 (using US SP ammo) 3030/8mm/ect.

I favor the .30 cal's with 150+ gr. bullits because I think they kill clean and knock down these big deer.  Don't have to be moa or traveling at the speed of light, just a big bullit to turn out the lights.  The biggest problem is getting one in your sights, that is finding a place to hunt and then having patience waiting for one to walk by, not get buck fever.  Our ancestors pretty much wiped out the US deer pop with black powder.  The truth is that most of us here, myself most of all, are gadget holics.  We need another gun like we need a dose of VD. 
P.S.  I just ordered another gun last night, go figure.

I'll go anywhere in the country and hunt nearly anything with a .308.... Most especially for deer.

I had an old 96 swede custom built for deer hunting this year, in what I feel is perfect for deer. The .250-3000 Savage.

Where I hunt long shots might get out to 100yds. I use the 308 it work well for me.

Have shot deer with 308, 30-06, 270, 358 Norma Mag, and 300 Win Mag.  I have been only shooting with my 300 Win Mag for the last 4 years using handloads with 200gr Sierra SP and have no problems with this combination as long as I do my part.  I always try for flank shots through heart and lung area and have taken shots from 30yds to 360yds so far.  Meat lost hasn't been a problem with the flank shots that I have taken and I won't take a shoulder shot since I want to preserve as much meat from damage as possible.  I use the 300 Win Mag since I hunt in a stand that has infinate range possibilities with islands of trees and brush all around this stand and no pressure from outside hunters, (long shots with varible winds).

Only one?  That is a hard one.  If I could only have one, it would be the old warhorse...the 30-06 springfield.  But I do also like the .270 Win, 7mm-08 Rem, and the great 45-70 Gov't.  But you said fav so it has to go to the venerable 30-06.

For some reason I just haven't changed my mind enough on this thred. So, today I'd like the 7x57, great cartridge. But tomorow is another day and it's hard to beat a 280 Rem. I think I've mentioned 5 or 6 now and at one time or other each was the favor of the moment!

  For me , in deep cover eastern mixed woods..where I usually hunt, I have two rifles that work well for me..

  A Marlin 336C 30/30 and an H&R/NEF .44 Mag..
  If I need go longer..I have a Savage and a NEF that are both .223 satisfactory for me since I believe in picking my shots..no running targets etc.

   Until 2 years ago the only long guns allowed in this area were shotguns w/slug..I used an H&R 12 ga smoothbore..very effective at moderate ranges..but I really prefer rifles.
